Elle repose principalement sur un m\u00e9canisme appel\u00e9 \"diffusion facilit\u00e9e\" (Facilitated Diffusion), qui est assur\u00e9 par un groupe de transporteurs sp\u00e9cialis\u00e9s appel\u00e9s transporteurs de glucose (GLUT).<\/p>\n\n\n\n<p>En termes simples, le glucose \u00e9tant une grosse mol\u00e9cule polaire, il ne peut pas directement \"traverser la paroi\" de la bicouche lipidique de la membrane cellulaire comme l'oxyg\u00e8ne. Il doit d'abord se lier \u00e0 une prot\u00e9ine GLUT sp\u00e9cifique \u00e0 la surface de la cellule. Une fois li\u00e9e, cette prot\u00e9ine subira un changement de conformation (vous pouvez comprendre cela comme une d\u00e9formation physique) et, sans consommer directement d'ATP, elle \"transportera\" le glucose dans la membrane le long du gradient de concentration.<\/p>\n\n\n\n<p>Il existe toutefois des exceptions en fonction des besoins physiologiques de l'organisme. Dans des tissus tels que l'intestin et le rein, le glucose entre par le biais d'un transport actif secondaire (reposant sur la prot\u00e9ine SGLT), qui est typique d'un \"reflux\" et qui est entra\u00een\u00e9 par la diff\u00e9rence de concentration des ions sodium. Dans les cellules musculaires et adipeuses, ce processus est enti\u00e8rement \"r\u00e9gul\u00e9\" par l'insuline. Ce n'est que sur instruction de l'insuline que la cellule d\u00e9ploie le transporteur GLUT4 vers la membrane cellulaire pour recevoir le glucose.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Why_Glucose_Cant_Drive_In_For_A_Long_Time\"><\/span>Pourquoi le glucose ne peut pas \u00eatre transport\u00e9 pendant longtemps<span class=\"ez-toc-section-end\"><\/span><\/h2>\n\n\n\n<p>Pour comprendre ce m\u00e9canisme de transport, il faut d'abord examiner la \"paroi\", c'est-\u00e0-dire la membrane cellulaire. La membrane cellulaire est constitu\u00e9e d'une bicouche lipidique, qui est essentiellement une barri\u00e8re s\u00e9lective. Son noyau interne est extr\u00eamement hydrophobe.<\/p>\n\n\n\n<p>D'apr\u00e8s notre analyse des propri\u00e9t\u00e9s mol\u00e9culaires, le glucose veut entrer seul et se heurte \u00e0 deux obstacles de taille :<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Taille<\/strong>: En tant que mol\u00e9cule, elle est relativement trop grosse.<\/li>\n\n\n\n<li><strong>Polarit\u00e9<\/strong>: Le glucose est une mol\u00e9cule polaire, c'est-\u00e0-dire hydrophile, mais il est bloqu\u00e9 par le noyau lipophile de la membrane cellulaire.<\/li>\n<\/ul>\n\n\n\n<p>C'est en raison de ces propri\u00e9t\u00e9s que le glucose ne peut pas se diffuser facilement \u00e0 travers la membrane, comme l'oxyg\u00e8ne ou le dioxyde de carbone. Ce processus d\u00e9pend enti\u00e8rement de la famille des transporteurs de glucose (GLUT). On peut consid\u00e9rer ces transporteurs comme des canaux ou des vecteurs sp\u00e9cialis\u00e9s int\u00e9gr\u00e9s dans la membrane cellulaire.<\/p>\n\n\n\n<p>Ce flux de travail est tr\u00e8s similaire \u00e0 la logique que nous utilisons lors de la conception de vannes de pr\u00e9cision :<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Reliure<\/strong>: Le glucose flottant dans le liquide extracellulaire se lie \u00e0 un site sp\u00e9cifique sur la face externe de la prot\u00e9ine GLUT.<\/li>\n\n\n\n<li><strong>Changement de conformation<\/strong>: Lors de la liaison, la prot\u00e9ine GLUT est imm\u00e9diatement \"d\u00e9form\u00e9e\". Elle inverse son orientation - elle est ferm\u00e9e vers l'ext\u00e9rieur de la cellule et ouverte vers l'int\u00e9rieur de la cellule.<\/li>\n\n\n\n<li><strong>Lib\u00e9ration<\/strong>: Les mol\u00e9cules de glucose sont ensuite lib\u00e9r\u00e9es dans le cytoplasme.<\/li>\n\n\n\n<li><strong>Passif<\/strong>: Le point cl\u00e9 ici est que le processus lui-m\u00eame ne n\u00e9cessite pas d'apport direct d'\u00e9nergie (ATP).
