Si basa principalmente su un meccanismo chiamato \"diffusione facilitata\" (Facilitated Diffusion), eseguito da un gruppo di trasportatori specializzati chiamati trasportatori di glucosio (GLUT).<\/p>\n\n\n\n<p>In poche parole, poich\u00e9 il glucosio \u00e8 una molecola grande e polare, non pu\u00f2 \"attraversare\" direttamente la parete del bilayer lipidico della membrana cellulare come l'ossigeno. Deve prima legarsi a una specifica proteina GLUT sulla superficie cellulare. Una volta legata, questa proteina subisce un cambiamento conformazionale (si pu\u00f2 intendere come una deformazione fisica) e, senza consumare direttamente ATP, \"trasporta\" il glucosio nella membrana lungo il gradiente di concentrazione.<\/p>\n\n\n\n<p>Tuttavia, esistono delle eccezioni che dipendono dalle esigenze fisiologiche dell'organismo. In tessuti come l'intestino e il rene, il glucosio entra attraverso un trasporto attivo secondario (basato sulla proteina SGLT), che \u00e8 tipico del \"backwater\" ed \u00e8 guidato dalla differenza di concentrazione degli ioni sodio. Nelle cellule muscolari e adipose, questo processo \u00e8 completamente \"regolato\" dall'insulina: solo quando l'insulina lo richiede, la cellula dispiega il trasportatore GLUT4 sulla membrana cellulare per ricevere il glucosio.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Why_Glucose_Cant_Drive_In_For_A_Long_Time\"><\/span>Perch\u00e9 il glucosio non riesce a entrare per molto tempo<span class=\"ez-toc-section-end\"><\/span><\/h2>\n\n\n\n<p>Per comprendere questo meccanismo di trasporto, dobbiamo innanzitutto considerare la \"parete\", ossia la membrana cellulare. La membrana cellulare \u00e8 costituita da un bilayer lipidico, che \u00e8 essenzialmente una barriera selettiva. Il suo nucleo interno \u00e8 estremamente idrofobico.<\/p>\n\n\n\n<p>Secondo la nostra analisi delle propriet\u00e0 molecolari, il glucosio vuole entrare da solo e si trova di fronte a due difficili ferite:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Dimensione<\/strong>: Come molecola, \u00e8 relativamente troppo grande.<\/li>\n\n\n\n<li><strong>Polarit\u00e0<\/strong>: Il glucosio \u00e8 una molecola polare, cio\u00e8 idrofila, ma viene bloccata dal nucleo lipofilo della membrana cellulare.<\/li>\n<\/ul>\n\n\n\n<p>\u00c8 a causa di queste propriet\u00e0 che il glucosio non pu\u00f2 diffondere facilmente attraverso la membrana come l'ossigeno o l'anidride carbonica. Questo processo dipende interamente dalla famiglia dei trasportatori di glucosio (GLUT). Questi trasportatori sono canali specializzati o vettori incorporati nella membrana cellulare.<\/p>\n\n\n\n<p>Questo flusso di lavoro \u00e8 molto simile alla logica che utilizziamo nella progettazione di valvole di precisione:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Rilegatura<\/strong>: Il glucosio che galleggia nel fluido extracellulare si lega a un sito specifico sul lato esterno della proteina GLUT.<\/li>\n\n\n\n<li><strong>Cambiamento conformazionale<\/strong>: Al momento del legame, la proteina GLUT viene immediatamente \"deformata\". Inverte il suo orientamento: \u00e8 chiusa verso l'esterno della cellula e aperta verso l'interno.<\/li>\n\n\n\n<li><strong>Rilascio<\/strong>: Le molecole di glucosio vengono quindi rilasciate nel citoplasma.<\/li>\n\n\n\n<li><strong>Passivo<\/strong>: Il punto chiave \u00e8 che il processo in s\u00e9 non richiede un apporto diretto di energia (ATP).
